摘要:研究目的 本文主要探討RIT1在膠質瘤中的生物學功能及其相關的分子機制。研究方法利用數據庫對比分析RIT1在膠質瘤組織和正常腦組織中的表達及其與膠質瘤患者預后的關系;通過CCK-8實驗分析RIT1對膠質瘤細胞增殖的影響;通過流式細細胞儀檢測 RIT1對細胞周期的影響;通過Western blot檢測RIT1敲低或者過表達后P-AKT以及P-mTOR的表達變化。研究結果通過數據庫分析,結果表明RIT1 在膠質瘤中高表達并且與膠質瘤患者的預后負相關;CCK-8 法分析表明RIT1可促進膠質瘤細胞的增殖;流式結果顯示敲低RIT1,細胞G0/G1期比例增多,S期和G2/M期比例減少;而過表達RIT1,細胞G0/G1期比例減少,S期和G2/M比例增加;Western blot實驗結果表明敲低RIT1下調了P-AKT和P-mTOR的表達,而過表達 RIT1則上調了P-AKT和P-mTOR的表達。研究結論 RIT1在膠質瘤中是一個癌基因,并且可通過AKT/mTOR通路促進膠質瘤的增殖。

RIT1 promotes glioma proliferation via the AKT/mTOR pathway
Abstract:Objective To investigate the biological function of RIT1 in glioma and its related molecular mechanism. Methods Databases were used to analyze the differential expression of RIT1 in glioma patients samples and associated normal brian tissues, and its relationship with the prognosis of glioma patients. CCK-8 assay was used to detect the effect of RIT1 on glioma cell proliferation. Flow cytometry assay was used to analyze the effect of RIT1 on cell cycle. Western blot assay was used to detect the expression of P-Akt and P-mTOR after RIT1 silence or overexpression. Results Through database analysis, the results showed that RIT1 was highly expressed in glioma and it\'s expression was negatively correlated with the prognosis of glioma patients; CCK-8 assay showed that RIT1 could promote the proliferation of glioma cells. Flow cytometry showed that knockdown of RIT1 increased the proportion of G0 / G1 phase and decreased the proportion of S phase and G2 / M phase in glioma cells; Overexpression of RIT1 decreased the proportion of G0 / G1 phase and increased the proportion of S phase and G2 /M phase. Western blot showed that RIT1 silence downregulated the expression of P-Akt and P-mTOR, while RIT1 overexpression upregulated the expression of P-Akt and P-mTOR. Conclusion RIT1 is an oncogene in glioma and promotes the proliferation of glioma through the Akt / mTOR pathway.
